What is the pratical difference between a reliever who is an R2 and one who is rated R2 C0?

Definition of a "closer situation" - occurs whenever the defensive team has the lead and the tying run is at-bat or on-base from the 9th inning on.

If a game is in a "closer situation", ignore the R rating and use the C rating. If a pitcher doesn't have a C rating, they are automatically fatigued. This doesn't apply to the starting pitcher if he is in the game when the game goes into a closer situation.

More important than I thought. A R2 will automatically start at F0 in any closer situation. A R2/C0 will rather begin at F9, and then will go downward after that, but should finish the inning in the mid- or low-Fs
