To set a rotation or not

that is the question...
I like to manage my staff on a nightly basis so I rarely set a rotation. However if using a SP/RP for the purpose of super reliever is it best to set a rotation (leaving said SP/RP out of it) to ensure he can be used in the proper RP roles.
Also, if you set a rotation "in stone" can you still go in and set your staff up every night, and not effect the use of your super reliever (if he is excluded from the set rotation.
...and yes I did try to make that as confusing as possible.
