From the league rules:

2) Only those players who played at least one MLB game during the season for which the draft is being conducted may be selected.
3) If a player is drafted but then does not receive a card in the set for the coming season, he still remains on a team's roster. There are no replacement picks allowed later.
8) All owners must have a full 40 man roster upon completion of the annual draft.
9) [b:8ad4aeb413]There is no clock for round 1 of each annual free agent draft.[/b:8ad4aeb413] All subsequent rounds will operate with a 2-hour clock, as in the initial draft.

The draft does not "officially" start for a couple of days yet, however I think we can start whenever each manager is ready. NO CLOCK IN ROUND ONE.
