No. Any prospect that you intend to use in Season 1 of the league must have been promoted prior to the FA draft.
There is a loophole in the rules, as Roscodog mentioned. If you wait until AFTER the completion of the FA draft and then make an unbalanced trade, which leaves you with 29 or fewer rostered players, you may promote a prospect to fill that spot.
Excerpt from Prospect Rules 5 and 6:
5) Prospects
must be promoted prior to the spring free agent draft to be eligible to be rostered at the start of the first season of play
6)
After the spring free agent draft is complete, Prospects can be promoted to the major league roster at any time, provided there is roster space available.
The reason you must wait until AFTER the FA draft is complete is that you must finish the FA draft with 30 players on your roster. The spring free agent draft is not complete until all teams have 30 players rostered. Therefore, you could not open up a roster spot for a prospect promotion while the FA draft is in progress. Any roster spot opened up by a trade during the FA draft must be filled by a FA pick.
